Transferable Resistance a N D in Enterococci Aminoglycoside-modifying Enzymes

چکیده
Ten isolates of Streptococcus faecalis and two isolates of S. faecium were studied together with an NCTC strain of each species. Antibiotic susceptibility tests showed different patterns of high-level resistance to aminoglycosides. Three S. faecalis strains were highly resistant to gentamicin and other aminoglycosides. By means of a filter membrane technique, transfer of high-level resistance to aminoglycosides was demonstrated from S. faecalis to S . faecalis, from S. faecium to S. faecalis and from S. faecalis to S. faecium, the last of which has not previously been described. Aminoglycoside-modifying enzymes were assayed with radiolabelled cofactors. High-level resistance to gentamicin and other aminoglycosides could be attributed to the production of 2”-O-phosphotransferases, 3’-O-phosphotransferases, 6-0-adenylyltransferases and 6’-N-acetyltransferases. Other resistance mechanisms accounted for resistance in two strains of S. faecalis and one strain of S. faecium that were highly resistant only to streptomycin and one S. faecalis strain that was moderately resistant to all aminoglycosides. A low level of 6’-N-acetyltransferases was detected in the three strains of S. faecium but this did not confer high-level resistance to aminoglycosides and this trait could not be transferred.

Prevalence of Genes Encoding Aminoglycoside Modifying Enzymes in Clinical Isolates of Klebsiella Pneumoniae in the Hospitals of Borujerd
Background and Aims: Given the importance of aminoglycoside resistance in nosocomial and community infections caused by bacterial pathogenes such as Klebsiella pneumoniae (K. pneumoniae), the aim of this study was to determine the frequency of aac (6')- Ib and aac (3)- IIa, the genes encoding aminoglycoside modifying enzymes involved in aminoglycoside resistance. Material and Methods: A t...
متن کاملAnalysis of Aminoglycoside Modifying Enzyme Genes Responsible for High-Level Aminoglycoside Resistance among Enterococcal Isolates
Enzymatic modification results in high-level resistance to aminoglycoside (HLAR), which eliminates the synergistic bactericidal effect of combined exposure to a cell wall-active agent and an aminoglycoside. So aim of the study was to determine prevalence of HLAR enterococcal isolate and to study distribution of aminoglycoside modifying enzyme genes in them. A total of 100 nonrepeat isolates of ...

This study highlights the prevalence of aminoglycoside-modifying enzyme genes and virulence determinants among clinical enterococci with high-level aminoglycoside resistance in Inner Mongolia, China. Screening for high-level aminoglycoside resistance against 117 enterococcal clinical isolates was performed using the agar-screening method. Out of the 117 enterococcal isolates, 46 were selected f...

Mating experiments have shown that high-level resistance (minimal inhibitory concentration greater than 2,000 microgram/ml) to streptomycin and kanamycin, and resistance to penicillin-streptomycin and penicillin-kanamycin synergism are transferable by conjugation from resistant clinical isolates of enterococci to a sensitive recipient strain. Cesium chloride-ethidium bromide ultracentrifugation...
